We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation conditions should be determined by the user. High temperature treatment of formalin-fixed tissue sections with 10mM citrate buffer, pH 6.0 must be performed prior to the immunostaining. Specificity: This antibody reacts with a 116 kD protein, known as PARP. Proteolysis of PARP to its stable 85 kD fragment is an early marker of apoptosis and is mediated by Caspase (CPP32) protein. PARP uses nicotinamide adenine dinucleotide (oxidized form) as a substrate to catalyze the transfer of ADP-ribose to a variety of nuclear acceptors. This antibody cross reacts with human, mouse and rat. Cellular Localization: Nuclear.
